چکیده

The importance of dexamethasone is demonstrated by its wide uses in human and veterinary medical practices [14]. Clinically, dexamethasone is administered to animals for the treatment of disease conditions such canine distemper, meningitis, necrotizing enterocolitis [5-10]. In humans, dexamethasone is most notably, used for the treatment of various complications, such as preterm complications to stimulate surfactant production by the lungs [3,11], intrahepatic cholestasis [12] and inflammatory conditions during pregnancy [13]. These actions are critical to prepare the fetuses for extrauterine life. Although this treatment greatly improves fetal and neonatal survival [3,14], they are not without adverse effects. Dexamethasone use has been associated with intra uterine fetal growth restriction (IUGR), decrease birth weight and placental weights in some animal models as well as humans [15-22]. The mechanisms underlying inhibitory effects of dexamethasone have largely not been elucidated. However, dexamethasone may probably exert its effect through fetal fluid medium. Amniotic and allantoic fluids protects the fetus by forming a protective physical cushion around it and allows the developing fetus to move freely inside the uterus and prevent teratogenicity due to adhesion and maintains an environment of constant temperature [23-25]. Fetal fluids are usually produced by placenta and other placentomes or fetal membrane during gestation until kidneys take over the task [23,24]. Other sources of fetal fluids include secretions from amniotic epithelium, fetal saliva, and nasopharyngeal secretions and skin, excretions of fetal kidneys and allantoic epithelia [26]. The biochemical constituents of amniotic fluid and allantoic usually provide valuable information for the assessment of fetal physiology and Abstract

Intergenerational consequences of fetal programming by in utero exposure to glucocorticoids in rats.

Epidemiological studies linking low birth weight and subsequent cardiometabolic disease have given rise to the hypothesis that events in fetal life permanently program subsequent cardiovascular risk. The effects of fetal programming may not be limited to the first-generation offspring. We have explored intergenerational effects in the dexamethasone-programmed rat, a model in which fetal exposur...

The comparative clinical efficacy of intravenous insulin regular, dexamethasone and flunixin meglumine on ovine experimental endotoxemia

The lack of an effective treatment for endotoxemia causes high mortality rates in affected animals. Forty clinically healthy 1-year old Iranian fat-tailed ewes were assigned randomly into 8 experimental equal (n = 5) groups, comprising Insln 1.5, Insln 3, Insln 6, Insln 9, Insln 20, Dexa, Flnx and Control. Lipopolysaccharide from E. coli serotype O55:B5 was used to induce endot...

Regulation of expression of the gene encoding the major surfactant protein (SP-A) in human fetal lung in vitro. Disparate effects of glucocorticoids on transcription and on mRNA stability.

We previously observed that dexamethasone had a biphasic effect on the levels of mRNA encoding the major surfactant protein (SP-A) in human fetal lung in vitro; at concentrations of 10(-10) and 10(-9) M, dexamethasone caused an increase in the levels of SP-A mRNA, whereas at concentrations of greater than 10(-8) M, the steroid had a pronounced inhibitory effect on SP-A mRNA levels. It was also ...

Background and Aim: One of the proposed ways for enhancement of labor induction is administration of corticosteroids. Although animal studies have shown the importance of cortisole in the initiation of labor, the role of corticosteroids in enhancement of labor process is still uncertainThe aim of the present study was to determine the effects of intramuscular dexamethasone on labor duration...

Dual effect of dexamethasone administration on anxiety and depression in adult male rats

Introduction: Dexamethasone is a synthetic glucocorticoid with possible effects on anxiety and depression because it has direct effects on the hypothalamus-pituitary-adrenal (HPA) and interacts with several neurotransmitter systems such as GABA and glutamate. Methods: Levels of anxiety and depression in rats were measured 40 minutes and 24 hours, respectively, after subcutaneous injection o...
